Description
Description: This 3D model represents a diagnostic Immunoglobulin G (IgG) antibody conjugated to the alpha subunit of beta-galactosidase, specifically the N-terminal fragment spanning residues 1-77 (the Enzyme Donor). In the ESSENCE nanoplatform, this alpha-donor fragment is an inactive polypeptide that restores full enzymatic activity only upon binding with its larger complementary partner, the omega-acceptor (the M15 deletion mutant). Technical Significance: Complementation: The 1-77 fragment acts as the essential key that restores the structural integrity of the enzyme active site and stabilizes the tetrameric state. Readout: Reconstitution of the functional enzyme triggers the hydrolysis of colorimetric substrates, enabling the wash-free detection of skin cancer biomarkers (IL-1alpha or CTSK). Prototyping: This model allows for the precise spatial mapping of the enzyme donor within the vertical channels of the diagnostic patch. Structural Note: This model is rendered as a high-fidelity surface mesh to illustrate the relative scale between the large IgG scaffold (150 kDa) and the smaller alpha-peptide donor (8.5 kDa).
